Raków were paired with Italian side Fiorentina, while Lech will take on Ukraine’s Shakhtar Donetsk.
The first legs are scheduled for March 12, with the return matches a week later.
Lech, who advanced via the playoff round, will host the first leg.
Raków, who qualified directly for the round of 16 after finishing second in the league phase, will play their opener away.
Because of the war in Ukraine, Shakhtar are playing their home Conference League matches this season in Kraków, southern Poland, at the municipal stadium normally used by Wisła Kraków.
In October, they lost 1-2 there to Legia Warsaw in the league phase.
If Raków eliminate Fiorentina, they will face the winner of the tie between Crystal Palace and Cypriot side AEK Larnaca in the quarterfinals.
Should Lech get past Shakhtar, they will meet either Dutch club AZ Alkmaar or Sparta Prague.
The bracket is structured in such a way that the two Polish sides could meet in the semifinals.
Lech reached the round of 16 by defeating Finland’s KuPS Kuopio 2-0 away and 1-0 at home in the playoffs.
Another Polish club, Jagiellonia Białystok, were knocked out by Fiorentina in the playoffs, losing 0-3 at home before winning 4-2 after extra time in Italy.
Legia Warsaw failed to progress from the league phase.
Fiorentina are two-time Conference League runners-up, losing the 2023 final to West Ham United and the 2024 showpiece to Greece's Olympiacos. They reached the semifinals last season.
The Conference League final is set for May 27 in Leipzig, Germany.
Europe’s third-tier club competition has become an important source of ranking points and prize money for Polish teams.
Thanks largely to strong performances by Legia and Jagiellonia, who both reached the quarterfinals last season, five Polish clubs will compete in European competitions in 2026-2027 for the first time, including two in Champions League qualifying.
